The seminar is followed by a lunch (kindly provided by the department) in order to allow everybody to interact and promote collaboration between labs.

12:00 - “Overexpression of human wild-tape APP alters GABAergic transmission” - Anna Kreis (FYCL lab)

Abstract

Mutations, overexpression or altered processing of the amyloid precursor protein (APP) are involved in the progression of Alzheimer’s disease (AD), characterized by irreversible loss of cognition and memory due to formation of extracellular amyloid plaques (Aβ) and neurofibrillary tangles (NFTs). Synaptic dysfunction leading to atypical neuronal synchronicity and oscillations appear in AD brain long before the characteristic Aβ and NFT deposits. APP and its cleavage products are reported to interact with receptors involved in synaptic release machinery therefore suggesting direct involvement in neurotransmission. Previous observations showed a downregulation of the K+-Cl- co-transporter (KCC2) in post-mortem AD brains and that overexpression of APP in neuronal culture induces a decrease of the chloride extruder KCC1. Therefore suggesting a possible change in a chloride transmembrane gradient disrupting GABAergic inhibitory transmission. Using a rarely utilized mouse model overexpressing human wild-type APP without the presence of amyloid plaques, we aim to unravel the importance and influence of APP on inhibitory and excitatory neurotransmission.
