Our team (ContrasT Team), under the supervision of Prof. Ir. Greet Kerckhofs, focuses on contrast-enhanced X-ray microfocus computed tomography (CECT) as an emerging 3D histology tool that enables to visualize and structurally analyze multiple biological tissues in full 3D at the same time. For now, sectioning followed by colorimetric or fluorescent staining is still the gold standard for histological analysis of tissues. However, it is well known that this technique has limitations with regards to the 3D microstructural assessment of biological tissues and their constituents, and to the quantification of their spatial interactions. It is also a destructive, labor-intensive technique, and is mostly restricted to 2D sections. Using contrast-enhancing staining agents (CESAs), CECT can tackle these shortcomings, allowing soft tissue visualization in full 3D. The ContrasT Team attempts to develop, optimize and validate novel CESAs and staining protocols. Within this context, my postdoc project focuses on the development of antibody-based CECT staining protocols for whole tissues, similar to immunohistochemical stainings on tissue sections.
